There are over 10,000 species of birds in the world, and it has been estimated that about half of them are sexually monomorphic, meaning that one cannot distinguish a male from a female just by looking at them. Although monomorphic males and females of the same species often show behavioral differences, such visual judgement of gender is not reliable, and certainly cannot be used in young, immature birds. For several decades, a surgical method (involving endoscopic inspection of internal organs by a veterinarian in a bird under anesthesia) has been used for sexing, and was the method of choice.

The sex in birds (and any other vertebrates) is genetically determined, and with the development of new technologies it is now possible to identify sex-specific markers for chromosomes that differentiate between males and females: the males have two identical Z chromosomes, while females have one Z and one W chromosome.

During the last 15 years, very sensitive techniques were developed allowing the detection of markers which are diagnostic for either sex chromosome in many avian species. During the last decade, DNA sexing has become very popular among breeders of exotic birds (which are often sexually monomorphic), and also finds its applications for preservation and planned breeding of endangered species. As the DNA of the same bird is identical in all cells of its body, one can use any "live" material for the tests. Because of its non-invasiveness, the preferred materials are pulled feathers, or a dried drop of blood collected from a clipped toenail. We can also use the eggshell from a newly hatched chick. In this case, the source of DNA is not the eggshell itself, but rather the membrane stuck to it on the inside.
